If the rims and tire are designed for tubeless systems then it will work.
I have tubeless on my 29 Plus bike (non thorn obviously) and it has worked flawlessly so far. The rims are WTB Scraper and the tires Vittorio Bomboloni. It was professionally set up, I didn't do it. Finished the last part of the Great Divide Mountain Bike Route on it. Encountered many goathead and other thorns, punctures, but no flats. They all sealed. My riding buddy had four or five flats.
Having said that, I see no need to convert my other bikes to tubeless, but with the right gear it works.
